Título: Development of a study protocol for regulatory testing to identify endocrine disrupting substances in biotic systems
Resumen:El proyecto Integrated Fish Endocrine Disruptor Test (iFEDT) se diseñó para establecer un nuevo sistema de ensayo que combina/amplía dos ensayos existentes en peces: el ensayo de reproducción a corto plazo en peces (OCDE TG 229) y el ensayo de desarrollo sexual en peces (OCDE TG 234). El propiltiouracilo (PTU) y el etinilestradiol (EE2) se probaron en peces cebra como sustancias modelo para la alteración del sistema hormonal tiroideo y sexual, respectivamente. Además de los criterios de valoración establecidos por las directrices técnicas existentes (fecundidad, crecimiento, histopatología gonadal, vitelogenina), se evaluaron los criterios de valoración relacionados con la tiroides, como los niveles hormonales, la histopatología tiroidea y el desarrollo (ocular) durante la exposición de 85 días.
Summary:The Integrated Fish Endocrine Disruptor Test (iFEDT) project was designed to establish a new test system that combines/extends two existing fish tests: the Fish Short Term Reproduction Assay (OECD TG 229) and the Fish Sexual Development Test (OECD TG 234). Propylthiouracil (PTU) and ethinylestradiol (EE2) were tested in zebrafish as model substances for disruption of the thyroid hormone and the sexual hormone system, respectively. In addition to endpoints set by existing TGs (fecundity, growth, gonad histopathology, vitellogenin), thyroid-related endpoints such as hormone levels, thyroid histopathology and (eye) development were assessed during the 85-d exposure.
