This increases the risk of high blood pressure and atherosclerosis which can lead to heart attacks, strokes and other diseases of the vascular or circulatory system.<\/p>\n<p style=\"text-align: justify;\">\u201cWe still don\u2019t have a good therapy to prevent the consequences of ageing or vascular ageing, such as hypertension,\u201d says project coordinator Judith Sluimer, professor of Cardiovascular Pathophysiology at <a class=\"link--external\" href=\"https:\/\/www.mumc.nl\/en\" target=\"_blank\" rel=\"noopener noreferrer\">Maastricht University Medical Centre<\/a> in the Netherlands.<\/p>\n<p style=\"text-align: justify;\">\u201cThere are treatments, but there are still a lot of people having heart attacks and strokes. So we need something better.\u201d<\/p>\n<p style=\"text-align: justify;\">Most research on ageing and atherosclerosis that looks at the inside of the blood vessel, studies its inner layers, Sluimer notes. \u201cThere are three blood vessel layers. Most people are studying the inner and the medial ones. So if we have new information on the cells in the outside layer, maybe we can find a new therapy,\u201d she adds.<\/p>\n<p style=\"text-align: justify;\">Her own expertise is in the effects of ageing on the adventitia, the outer layer. Her laboratory is also involved in the EU-funded <a class=\"link--external\" href=\"https:\/\/www.era-cvd.eu\/\" target=\"_blank\" rel=\"noopener noreferrer\">ERA-CVD<\/a> (Cardiovascular Disease) Consortium\u2019s MEND-AGE project.<\/p>\n<h6 class=\"c-article__title\"><span style=\"font-size: 14pt;\">Studying the function of fibroblasts<\/span><\/h6>\n<p class=\"c-article__text \" style=\"text-align: justify;\">Research undertaken by Dlzar Kheder at Maastricht University Medical Centre in the FIB-AGE project, which was funded by the <a class=\"link--external\" href=\"https:\/\/ec.europa.eu\/research\/mariecurieactions\/node_en\" target=\"_blank\" rel=\"noopener noreferrer\">Marie Sk\u0142odowska-Curie Actions<\/a> programme, looked at the function of collagen-producing fibroblasts in the outside layer and how it relates to changes that occur with ageing and cardiovascular disease.<\/p>\n<p class=\"c-article__text \" style=\"text-align: justify;\">\u201cFibroblasts are a type of cell that normally only live in the outside layer on the vasculature. But they also live in many organs,\u201d Sluimer explains. \u201cThey are interesting cells because they are known, for instance, for regeneration and wound healing of the skin.\u201d<\/p>\n<p class=\"c-article__text \" style=\"text-align: justify;\">\u201cFibroblasts also stimulate the formation of tiny new blood vessels to help regenerate tissue,\u201d she adds.<\/p>\n<h6 class=\"c-article__title\" style=\"text-align: justify;\"><span style=\"font-size: 14pt;\">Identifying markers<\/span><\/h6>\n<p class=\"c-article__text \" style=\"text-align: justify;\">To study the function of the cells, the project had identified some adventitial cell markers to distinguish them from the medial cells.<\/p>\n<p class=\"c-article__text \" style=\"text-align: justify;\">\u201cUsing single-cell sequencing, we looked at the transcriptomes of the mRNA expression of individual cells, one by one, in order to validate adventitial fibroblast-specific markers for these cells. At that point, nobody knew how to distinguish fibroblasts from medial cells,\u201d Sluimer explains.<\/p>\n<p class=\"c-article__text \" style=\"text-align: justify;\">Then, a knockout mouse \u2013 a rodent that had been genetically engineered to switch off the specific marker gene \u2013 was shipped to the Netherlands from Canada. They found the knockout had higher blood pressure, showing that the particular marker potentially has a role in causing hypertension.<\/p>\n<p class=\"c-article__text \" style=\"text-align: justify;\">Kheder, who was also part of the EU-funded <a class=\"link--external\" href=\"https:\/\/vascagenet.eu\/\" target=\"_blank\" rel=\"noopener noreferrer\">VascAge<\/a> network for research into vascular ageing, used his expertise culturing segments of arteries in an organ bath ex vivo to study the contraction and relaxation of isolated blood vessels, comparing their function in knockout and wild mice.<\/p>\n<table style=\"width: 99.9988%; border-collapse: collapse; border-style: double; border-color: #001f7d;\" cellpadding=\"0,5\">\n<tbody>\n<tr>\n<td style=\"width: 100%;\"><a href=\"https:\/\/www.cde.ual.es\/la-genetica-y-su-influencia-en-el-peso-alto-o-bajo-al-nacer\/\" target=\"_blank\" rel=\"noopener\"><img decoding=\"async\" class=\"alignleft\" src=\"https:\/\/www.cde.ual.es\/wp-content\/uploads\/2021\/02\/photo-1544126592-807ade215a0b.jpg\" alt=\"\" width=\"201\" height=\"134\" \/><\/a><\/p>\n<h4 class=\"entry-title\"><a href=\"https:\/\/www.cde.ual.es\/en\/genetics-and-their-role-in-small-and-large-birth-weight\/\" target=\"_blank\" rel=\"noopener\">Genetics and their role in small and large birth weight<\/a><\/h4>\n<\/td>\n<\/tr>\n<\/tbody>\n<\/table>\n<h6 class=\"c-article__title\" style=\"text-align: justify;\"><span style=\"font-size: 14pt;\">Role in human hypertension<\/span><\/h6>\n<p class=\"c-article__text \" style=\"text-align: justify;\">In parallel, the project worked on gene function. \u201cWhat we are trying to show is, can we rescue the accelerated ageing that we see in the knockout,\u201d Sluimer says. \u201cThen see if humans with this DNA variant have a lower amount of the gene as well, and whether we can rescue either the gene or the function of the gene to prevent hypertension occurring.\u201d<\/p>\n<p class=\"c-article__text \" style=\"text-align: justify;\">One of the genes had one of those DNA variants with hypertension, Sluimer notes, adding it was important proof that, in humans too, without the fibroblast gene there is increased hypertension.<\/p>\n<p class=\"c-article__text \" style=\"text-align: justify;\">But she remarks: \u201cWe still have a long way to go, because we don\u2019t know yet how to translate that into a therapy.\u201d<\/p>\n<p><strong>More information: <\/strong><a href=\"https:\/\/cordis.europa.eu\/article\/id\/447096-can-ageing-be-slowed-research-on-fibroblasts-may-show-the-way\" target=\"_blank\" rel=\"noopener\">CORDIS<\/a><\/p>\n","protected":false},"excerpt":{"rendered":"<p>A novel approach to studying fibroblasts \u2013 cells that are important for the formation of connective tissues \u2013 could be a route to new therapies for atherosclerosis and other diseases of the vascular system.
